When I use the Model master red putty or the Squadron green or white putties, I let them dry overnight just to be on the safe side. The Squadron stuff seems to take a little longer to cure than the Model Master stuff though, I don’t know why. I’ve recently read about using liquid paper to cover over small gaps. So far I haven’t had any problems with the liquid paper. If you decide to try it, let the part set for about 10-15 minutes before sanding.

It completely depends on how thick the layer of putty is, and the humidity of the room. Thin layer in a low humidity room means a quicker dry time.
Personally, I've dumped all the modeling putties and always use Bondo Spot Putty. Much cheaper and works better. In a thin layer it can be ready to sand in half an hour.
